GHK-Cu was found to reset gene expression patterns in aged human skin fibroblasts toward a younger state — activating over 4,000 genes involved in collagen synthesis, anti-inflammatory response, and tissue repair. The authors identified it as one of the most potent naturally occurring skin-repair signals known.

Applied topically, GHK-Cu significantly increased collagen production in human dermal fibroblasts in vitro. The study confirmed the peptide's ability to stimulate type I and type III collagen synthesis — the structural proteins responsible for skin firmness and elasticity — even at low concentrations.

In a double-blind clinical trial, subjects using a GHK-Cu complex showed statistically significant improvements in fine lines, skin laxity, and overall skin appearance versus placebo. Improvements were observed in mottled pigmentation and skin density after 12 weeks of consistent topical use.

A 12-week split-face clinical study found that 5% topical niacinamide significantly reduced hyperpigmentation, fine lines, skin sallowness, and pore size compared to vehicle control. Niacinamide was also shown to increase ceramide levels in the skin, directly strengthening the barrier function.

This study demonstrated that topically applied Hyaluronic Acid — particularly low molecular weight forms — penetrates the epidermis and significantly increases skin hydration, smoothness, and elasticity. After 8 weeks, subjects showed measurable reductions in wrinkle depth and significant improvements in skin firmness.

Pickart's foundational dosing research established that GHK-Cu exerts its strongest regenerative effects at concentrations in the range of 1–10 mg/mL, with optimal tissue-remodeling activity observed around 5 mg/mL. Below 1 mg/mL, biological effects were markedly reduced — which is why most commercial serums dosed under this threshold show limited real-world results.
